Routine 3D cell culture has arrived

 

 

Example application: Culturing primary hepatocytes in 3D

 

Liver hepatocytes are often used to test drug toxicity in vitro. There is good evidence which shows that liver cells grown in 3D cell culture out perform their 2D counterparts. Below shows SEM images (left and middle) of hepatocytes grown on different formats of polystyrene substrate, namely 2D conventional plasticware and 3D alvetex® scaffolds. The TEM (right) shows the formation of a bile cannaliculus and presence of tight junctions between cells growing in 3D.

Improved primary hepatocyte toxicity screens using alvetex®

The survival rate of primary liver hepatocytes in vitro is poor. reinnervate has shown that growing primary hepatocytes on 3D polystyrene scaffolds increases cell viability significantly compared to growth in standard tissue culture plastic.

Cytochrome P450-1A2 induction in primary rat hepatocytes is significantly elevated in cells grown in 3D polystyrene scaffolds compared to the 2D controls. Cells were induced to express cytochrome P450 using 3-methylcholanthrene. Also shown are non-induced controls.
