The LGC Standards Cell Line Authentication report has been formatted to meet the needs of cell biologists.
STR profiling is a forensic science which has been designed specifically for identifying an individual person
from within a population. It is the same technology which is used to identify perpetrators of crime from
samples taken at the scene. Kits designed for this forensic work will also create profiles for cell lines of
human origin.
STR profiling involves amplifying small sections of genomic DNA which are found at specific loci.
Separation of these amplicons and mapping them to a size standard creates the profile. Fresh samples
taken from an individual generally yield a good quality profile, where there is low background, and the profile
shows either a homozygote or heterozygote at any given loci and ideally the dual peaks of a hetrozygote
are balanced, with both similar peak height and area under the peak.
Cell lines are unusual. Many are from primary or metastisised cancers, others are combinations of 2 lines
which are fused, and some have been artificially immortalised. The unusual genetic make up of these cells
means that it is common to see significantly unbalanced hetrozygotes, triple peaks at a particular loci- many
of which are unbalanced, or for some lines it is normal to have 5 individual peaks at a single loci. Interpreting
the information from your cell line’s STR profile can be a little daunting.
LGC Standards have developed a cell line authentication report which provides you with a clear result for
each of your samples. You receive a single page per sample which includes:
peak calls for your cell sample;
reference profile for your cell line (where available);
EPG image showing the position of the peaks;
comments on the profile;
recommendations on next actions;
where the sample has not matched the reference provided, the most likely identity.
Also you will receive copies of EPGs as individual images that you may include in your report or publication
appendix.
For ease we include a covering results page which allows you to identify which samples have passed and
to begin an investigation of any which have returned aberrant data.
Many providers provide you with only the peak calls leaving you to work out the rest. We don’t. Our data
